There are some interesting ideas at the bottom of this article ("The Perfect Direct Mail Piece in Twelve Easy Steps):

I like the "tabloid headline" suggestion. You could really have some fun with that one.

Since all regions/demographics respond to marketing mediums in a different manner I would test both. Though a postcard may be better, it will cost money to design and print whereas a letter will simply require a letterhead and standard printing.

I would try this... Instead of doing 5000 postcards or letters, try doing 2,500 letters and 2,500 postcards. Put different contact info or some sort of coupon code on each. You will then be able to track where the leads came from.

    I hope that helps.

Letters are annoying as a potential client it irritates me to get letters that offer sale pitches.
I would rather see what you got without the cover up. Make it fancy and clear, not too much writing.

Regarding 4 x 6 versus 5 x 7 and 4 x 9 postcards, check your mail next time and if it is in a pile you will probably see the larger items at the back of the pile as this is how letter carriers sort their mail. I have read where people read from the largest to the smallest items.
