Try creating a email with a personal touch welcoming them to your site. Users may feel less pressure and they may comeback which is what you want and they may like getting a "personal" auto responder.

That's a good question. I get about 10 registrations a day and we have tried everything from personalized emails to simple welcome aboard emails. I am trying a new approach in the last few weeks with the goal of pointing the person who registered to what we want them to do when they use our site. here is what we send...
Hi (name),
Have you selected any favorite homes that you want to see in person? Did you know that you can schedule a tour direct from your new account on (our wesbite)? Simply click the “Request a Showing” button online and a Realtor from Great Austin Properties will contact you immediately to arrange the tour!

I try to send that email within a few hours of a new registration and we know it's already worked in a few instances. A few days later, we'll look at thier favorite homes and write a more personalized email about the homes they've chosen and again, offer to go see homes.

My first email is very polite and asks an obvious question. The whole thing looks like this:
"""
Hi (name),
Thanks for signing up at my site www mysite.com.
Would you like to buy a house?
(simple sig block to close with proper link back to website used to sign-up)
""
Notice that after "www" there's no period. Consequently, the chances of your email being treated like spam goes down significantly.
It's amazing how many times a terse, non-threatening question can start a long dialogue...

Our first letter introduces our Agent that will be helping them buy or sell a home and let's them know what we will be doing for them. We keep it two small paragrahs. All our other Drip letters are 1 paragraph. Keep it simple and relevant.

I usually just send out some type of follow up giving them user/pass with brokerage contact information and quick personal note like....if you see any properties you need details on or would like to view, please give us a call.

I think the auto-reply emails that go out after a sign up are junk and meaningless. I know gurus are out their professing that you need to respond ASAP, but that is not the answer.
I would rather receive a personal email 24 to 48 hours later than I would an inpersonal auto-reply. Anyone can tell when the response is just an auto-reply.
Keep it simple. Respond to their first email personally, and prompt them for some form of action (i.e. go to your website to gain access to "Up-to-the-Minute New Properties List", or to download a 2 page white paper on "____".)
Engage them and they will respond.
You could use an auto-reply with something like this "You requested some info from my site...great! I know this is just an auto-reply message but I wanted to provide you with some additional information that could be useful to you - and then of course I will respond to you personally by tomorrow."
auto replies are not meaningless if you offer something of value to the user, such as User/Pass and direct access page into your MLS database. This is usually the email user saves and refers to later when trying to search properties again.
i don't have my agents contact client until they personally request assistance or a showing on a particular property.
