Search engines cannot read video content as yet except, for some basic things like detecting adult content, etc. They rely on the meta tags, title, transcript, inbound links, anchor text and I believe even user comments to decide what the video is about.

Video on your site will not affect your Google Pagerank in any way. It can only serve to enhance user experience.

However, it can bring you lots of traffic. How? Just search for any keyword in Google and you will see video results on the first page mostly from sites like youtube or metacafe. This is a great way of jumping above your competition and being listed on the first page of organic search results even for highly competitive keywords. And if you submit to youtube, the same video can be embedded on your site too.

Secondly, Youtube itself is a very high traffic site. You have to use the 62 character limit of description and tags very wisely to siphon off Youtube traffic and get them to your website.

Thirdly, leads coming from a video sharing site are extremely qualified ones. Why? First, they searched and viewed your video. Second, they have to type your domain name in the browser to get to your site. That makes it a really extremely qualified lead.
